Gefitinib is an oral small molecule EGFR inhibitor that is approved for the therapy of metastatic non-small cell lung cancer. Side effects are usually dose related mild to moderate, with approximately 1% probability for intestinal lung disease (ILD) that is the most adverse event, while common chemotherapy-associated effects like myelosuppression and alopecia are not detected [62]. Gefitinib lowered ACTH synthesis in vitro and ACTH levels and corticotroph tumour size in AtT-20 corticotroph tumour xenograft mice in vivo [59]. Gefitinib’s antisecretory action in primary cultures of human corticotroph tumours was significant only in USP8 mutant cases [63]. Accordingly, a clinical trial is registered for the targeted therapy with gefitinib in patients with USP8-mutated CD, but no results were reported so far (ClinicalTrials.gov no. NCT02484755).
